Human/Rodent Somatic Cell Hybrid Mapping Panel #1

Mapping Panel #1 consists of DNA isolated from 18 human/rodent somatic cell hybrids retaining from one to 19 human chromosomes. Samples NA09925 through NA09940 were isolated from human/mouse somatic cell hybrids resulting from the fusion of fetal male human fibroblasts, IMR-91, with the TK-deficient mouse cell line B-82 and were submitted to the Repository by Dr. T.K. Mohandas, University of California, Los Angeles, UCLA-Harbor General Hospital. References describing the production and characterization of these hybrids include: R.T. Taggart, T.K. Mohandas, T.B. Shows, and G.I. Bell. Proc. Natl. Acad. Sci., USA 82:6240-6244, 1985 and T. Mohandas, C. Heinzmann, R.S. Sparkes, J. Wasmuth, P. Edwards, and A.J. Lusis. Somat. Cell Mol. Genet. 12:89-94, 1986.

Characterization of the above hybrids (see table) indicated a lack of representation by human chromosomes #9, 16, and X. The addition of DNA from three monochromosomal hybrids completed the panel.

Sample NA10611 was isolated from a human/Chinese hamster somatic cell hybrid submitted to the Repository by Dr. D. Warburton, Columbia University, New York, NY. This hybrid resulted from the microcell fusion of retroviral vector SP-1 infected human lymphoblast culture, GM07890, with a 46,XX,del(13) karyotype with Chinese hamster ovary line UV-135. The hybrid was selected in culture medium containing histidinol and retains an intact human chromosome #9. The production and characterization of this hybrid is described in: D. Warburton, S. Gersen, M.-T. Yu, C. Jackson, B. Handelin, and D. Housman. Genomics 6:358-366, 1990.

Sample NA10567 was isolated from a human/mouse somatic cell hybrid submitted to the Repository by Dr. D. Ledbetter, Baylor College of Medicine, Houston, TX. This hybrid, produced by Dr. D. Callen, Adelaide Children's Hospital, North Adelaide, Australia, resulted from the fusion of a human fibroblast cell line, GM02860, with a 46,XY,t(3;16)(q13.2;q13) karyotype, with the HPRT- and APRT-deficient mouse cell line A9. The hybrid was selected in culture medium containing aminopterin, adenine, thymidine, and L-proline and retains an intact human chromosome #16. The production and characterization of this hybrid is described in: Callen, Ann. Genet. 29:235-239, 1986.

Sample NA10324 was isolated from a human/mouse somatic cell hybrid submitted to the Repository by Dr. F. Ruddle, Yale University, New Haven, CT. This hybrid resulted from the fusion of a human fibroblast cell line, GM00144, with a 46,XX,-6,+der(6)t(6;21) karyotype with the HPRT- and APRT-deficient mouse cell line A9. The hybrid was selected in culture medium containing HAT. This hybrid retains an intact human X chromosome. The production and characterization of this hybrid is described in Dorman et al. Proc. Natl. Acad. Sci. USA 75:2363-67, 1978.

In addition to the human/rodent somatic cell hybrid DNA samples, Mapping Panel #1 includes DNA samples isolated from IMR-91 (NAIMR91), Chinese hamster ovary line UV-135 (NA10908), and mouse line B-82 (NA00347).

The Repository will maintain a database for results obtained with the panel which will be available to all investigators. It is not the Repository's responsibility to resolve discrepancies that investigators might find when using the NIGMS mapping panel.
